I’m sure someone else will chime in saying the main culprit is probably going to be the foam filter, will probably tell you one of three things, it’s getting too much air not enough gas at WOT, change your jets/carb, or reinstall a stock or similar airbox. There’s also some info you left out such hours/miles are on the rebuild that would help others pinpoint the problem without seeing the bike in person or a video to analyze.

The cylinder has about 800km on it and I put a new piston and rings in about 500km ago because the exhaust bolts broke off in the cylinder so I had it off

Last Wednesday the pressure line that pushes fuel out of the tank came off. The line might have some dirt in it so no pressure will be getting pushed through. The sputtering is like a struggle to rev out. When it’s held full throttle it cuts in and out like the kill switch is getting turned on and off, just like the sound it made when the pressure line came off. If we are talking about a vacuum hose (not fuel) that stretches from the carb to the fuel tank's valve - That line definitely must be clean and air-tight. If it came off, that might mean that it's not secured properly and some vacuum is escaping through it! Check for cracks and moisture. If there is any amount of water in the vacuum line that will definitely make the whole system malfunction. To test if there is a vacuum leak where the hose connects to the fuel tank, you should remove one end from the carb and blow into it. No air should be comming through and pressure should stay the same. After doing that remove the hose once again and check for any moisture. Also, if there is any water in the fuel tank's valve - dry it. I have had a simmilar issue before and this seemed to fix it.

Maybe the fuel valve (vacuum operated petcock) is no good. Could be leaking air into the fuel side or fuel into the vacuum side. Transparent fuel/vacuum hoses are very useful. I could also be that at high throttle positions, the vacuum that opens the petcock is not strong enough to keep the valve fully open, thus starving the fuel flow at higher throttle positions.
I have transparent fuel lines and I swapped out my vacuum petcock for a manual fuel shut off valve.
