I've got the Polini PWK in 24" size. I did my initial setup and have it about as good as I can get it, but I'm finding it hard to perfect the last 5%. If I place the needle clip at the top position, it runs a bit rough (rich) at 1/4 throttle but perfect up to WOT. If I place the needle one step down, it takes off even better, but it feels rich and 4-strokes at 3/4 throttle.
I look at this chart and can't figure if I should be looking for a different needle, or try to compensate by lowering the main jet. I just don't know how much the overlap impacts it.
Current settings are... Needle clip 2nd from top Main jet 110 Pilot 45

What size Main jet? What size Pilot jet are you running?

I was dealing with something similar on my Polini CP earlier this week. I was running great from idle to half, rich bog at 3/4, and great at WOT. I ended up going leaner on the needle and richer on the pilot. In your circumstance, I definitely think you could try leaning out the pilot and leaving the needle where it's at. Go a few sizes down and see where your idle screw has to be to get it to run right.

I posted above the jetting info. I still think it's a bit rich at 3/4 throttle(with pilot 45, main 110, needle 2nd from top). Just not sure if it's the needle or the main jet. I'm going to do some more testing with the main because it's running perfect up to 3/4 throttle. I think I've lost a touch at the very top so I'm guessing I need to lean out the main. I've noticed it's running much cooler then I used to. Like a drop of 50F, so I'm thinking it's very rich at the 3/4 to WOT.
I just haven't had time to test because it's always raining when I get out of work.

Since the bike is in a decent state of tune, i would:
--focus on Main Jet tuning first. That's what i do. I keep changing out main jets and go right into extended WOT runs. When i find a main jet that puts me at 370-350°, then i fiddle with mid throttle fine tuning.
