Well, from what I see, there is a a hose directly under there, if you look just to the left bottom of your circle, you see where the hose is connected to the carb.

You said that the hose I marked as DRAIN comes from the bottom of the carb? If so it needs to point down. The barb/ nipple should be where I have it circled. It could be on the manifold? Unlikely though.

-Yes confirm the red circled hose is located near the bottom of carb. -Is the nipple under that black hose you circled referring to? -is it possible to run vacuum fuel valve with vent and vac plugged.?

The nipple should be under that black wire (electrical supply to your auto enricher). No, the vacuum operated fuel valve must have vacuum in order to open. It must be connected. The vent can be capped (I think) but there's no reason to.
